چکیده

Abstract Kawasaki Disease (KD), a pediatric acute febrile illness and systemic vasculitis, is the leading cause of acquired heart diseases in children. Coronary artery aneurysms (CAAs) occur up to 25% untreated children, which reduced 5% with intravenous immunoglobulin (IVIG) treatment. However, 20% KD patients are refractory IVIG at higher risk CAAs, highlighting need characterize mechanisms mediating vasculitis develop more efficient therapies. IL-33 released upon inflammation tissue damage exerts its effects by binding receptor ST2 (IL1RL1). Levels elevated during phase. how contributes development cardiovascular lesions remains unknown. Using Lactobacillus casei cell wall extract (LCWE) murine model we observed increased Il33 mRNA expression hearts abdominal aortas LCWE-injected mice. While was detected immunofluorescence both control mice, mainly tissues from Single-cell RNA-sequencing mice indicated that transcripts were expressed primarily fibroblasts, endothelial cells, B vascular smooth muscle cells. In contrast, Il1rl1 highly infiltrating regulatory T macrophages, eosinophils, neutrophils. Blocking either anti-IL-33 antibody or using −/−mice significantly attenuated LCWE-induced vasculitis. Our results indicate promotes could therapeutically be targeted inhibit lesions.

JNK is critical for the development of Candida albicans-induced vascular lesions in a mouse model of Kawasaki disease.

BACKGROUND Kawasaki disease (KD) is the most common systemic vasculitis of unknown etiology in children, and can cause the life-threatening complication of coronary artery aneurysm. Although a novel treatment strategy for patients with KD-caused vascular lesions is eagerly awaited, their molecular pathogenesis remains largely unknown. c-Jun N-terminal kinase (JNK) is a signaling molecule known ...

Cholinergic neuropathology in a mouse model of Alzheimer's disease

Transgenic mice over-expressing mutant human amyloid precursor protein (PDAPP mouse) develop several Alzheimer’s disease (AD)-like lesions including an age-related accumulation of amyloid-?-containing neuritic plaques. Although aged, heterozygous PDAPP mice also exhibit synaptic and glial cell changes, that is characteristic of AD pathology, no evidence of neurodegeneration has been observed. T...

Elevated IL-33 expression is associated with pediatric eosinophilic esophagitis, and exogenous IL-33 promotes eosinophilic esophagitis development in mice.

We tested whether the T helper (Th) type 2 (Th2) cell agonist and allergenic ligand IL-33 was associated with eosinophilic esophagitis (EoE) development in a pediatric cohort and whether IL-33 protein could induce disease symptoms in mice. Biopsies from EoE patients or controls were used to measure IL-33 mRNA and protein expression. Increased expression of IL-33 mRNA was found in the esophageal...

Cardiovascular involvement in Kawasaki disease.

We report 72 patients with Kawasaki disease seen at this Centre over 7 years. Cardiac involvement in the form of mild pancarditis was seen in 28 % patients, but disappeared subsequently. Thirteen (18.5 percent) children developed coronary artery disease, out of which 4 resolved by the end of two months and another 6 after one year; 3 patients continued to show coronary artery dilatation and ane...
